The clip targets people who may be close to the person who was involved in her disappearance in 2007.
A Minute for Madeleine was released on Tuesday morning by the Child Exploitation and Online Protection (Ceop) Centre.
The film includes fresh images of how Madeleine might look now, aged six, including one with dark brown hair and tanned skin.
It has already been spread across thousands of social networking sites, blogs, and search engines. On Twitter, celebrities including Jonathan Ross, Chris Evans, Alan Carr and Phillip Schofield have "tweeted" the link to their followers.
Ceop Chief Executive Jim Gamble said: "We are delighted with the response so far but this is only the start."
Madeleine was nearly four when she went missing during a holiday in Praia da Luz in Portugal.
